Pedro Ponce estudió en el Colegio de Santa Cruz en México. Se graduó como licenciado en teología y, entre 1571 y 1626, tuvo a su cargo la diócesis de Zumpahuacán.
Una crónica de la época relata que: “En 1565 fue construido el templo parroquial para la congregación de los Agustinos … por orden de Pedro Ponce de León, probablemente hijo de Lucas Ponce de León, hijo del rey Cuatlatlapatzin de Tlaxcala”.
